DASRUL, MUSLIM AKMAL.Penelitian ini bertujuan untuk mengetahui pengaruh pemisahan spermatozoa dengan metode swim up terhadap kualitas spermatozoa, angka kebuntingan, dan rasio jenis kelamin anak kambing Peranakan Ettawa (PE). Pada penelitian ini digunakan 5 ekor kambing PE jantan berumur 18-24 bulan, yang ditampung masing-masing 1 kali tiap pejantan dengan menggunakan elektroejakulator. Semen berkualitas baik ditambahkan NaCl fisiologis 1:1 dan dibagi dalam 3 kelompok perlakuan. Kelompok 1 sebagai kontrol (P0) yaitu semen yang tidak dipisahkan dengan swim up. Kelompok 2 sebagai perlakuan 1 (P1) yaitu semen yang dipisahkan dengan metode swim up selama 5 menit. Kelompok 3 sebagai perlakuan 2 (P2) yaitu semen yang dipisahkan dengan metode swim up selama 10 menit. Parameter kualitas spermatozoa yang diukur adalah persentase motilitas, spermatozoa hidup dan abnormalitas spermatozoa. Sedangkan penilaian angka kebuntingan, dan rasio jenis kelamin anak diukur setelah perlakuan inseminasi pada induk betina. Data yang diperoleh dianalisis dengan analysis of variance (ANOVA) pola satu arah yang dilanjutkan dengan uji berganda Duncan. Hasil pengamatan konsentrasi, motilitas; spermatozoa hidup ; abnormalitas spermatozoa dan MPU spermatozoa setelah pemisahan berbeda secara nyata (P0,05) dibandingkan dengan swim up 10 menit. Demikian juga dengan angka kebuntingan dan rasio jenis kelamin kelahiran anak setelah pemisahan berbeda secara nyata (P0,05) dibandingkan dengan swim up 10 menit. Pemisahan spermatozoa dengan swim up dapat meningkatkan kualitas spermatozoa, angka kebuntingan dan rasio jenis kelamin anak jantan. Lama waktu swim up tidak berpengaruh secara nyata terhadap kualitas spermatozoa, angka kebuntingan dan rasio jenis kelamin anak jantan pada kambing PE. The purposes of this experiment are to investigate the separation of X and Y spermatozoa by measuring the spermatozoa quality, sex ratio between X and Y, capacity of fertility indicated by conception rate and sex ratio of goat boar kids. Samples, used in this experiment, are fresh semen from Boer goat with high quality consists of 4 group treatments with 6 replications 1) group of spermatozoa without separation (control), 2) group of spermatozoa separated by percoll gradient density centrifugation 3 levels (P1), 5 levels (P2) and swine up (P3). The observed parameters are spermatozoa quality, X and Y spermatozoa ratio, fertility’s capacity and sex ratio on the birth. Quality examination of spermatozoa and identify X and Y spermatozoa is based on the standard method of WHO. The conception rate was based on the ratio of pregnant goat after the first insemination. Data of spermatozoa quality and spermatozoa ratio were analyzed by using analisis of variance (ANOVA) and further analysis by LSD if there were differences between treatments. The results of this experiment showed that spermatozoa quality Boer goat significantly reduced (p<0,05) after separation with percoll gradient density centrifugation  and swim up. Percentage spermatozoa X after percoll gradient density centrifugation was significantly higher (P<0,05) compared to control and swim up. Meanwhile, the Y spermatozoa population was significantly higher (P<0,05) after swim up treatment compared to percoll gradient density centrifugation and control. The percentage of sex ratio (male: female) after insemination from percoll gradient density centrifugation produced more female than male. On the other hand, insemination from swim up produced more male than female. Sex ratio produced from separation of percoll gradient density centrifugation, swim up was difference from control semen (P<0,05). From this experiment, it was concluded that spermatozoa separation by percoll gradient density centrifugation and swim up can be used as one of the methods to separate   X and Y spermatozoa and further can be applied to get preferred sex animals. The research objective is fiercely attracting and challenging considering the similarity of bath physical and chemical properties among these elements. The rate separation of samarium and yttrium elements using supported liquid membrane has been studied. Polytetrafluoroethylene (PTFE) with pore size of 0.45 µm has been used as the membrane and di(2-ethylhexyl) phosphate (D2EHP) in hexane has been used as a carrier and nitric acid solution has been used as receiving phase. Result of experiments showed that the best separation rate of samarium and yttrium elements could be obtained at feeding phase of pH 3.0, di(2-ethylhexyl) phosphate (D2EHP) concentration of 0.3 M, agitation rate of 700 rpm, agitation time of 2 hours, and nitric acid and its solution concentrations of 1.0 M and 0.1 M, respectively.
