Dalam penelitian ini digunakan 20 ekor sapi yang dibagi menjadi dua kelompok, yakni kelompok sapi yang dilakukan restraint dengan restraining box tipe Mark 1 (K1, n=13) dan kelompok sapi yang dilakukan restraint dengan metode konvensional (K2, n= 7). Masing-masing kelompok diambil darahnya sebelum dan saat dipotong untuk diperiksa kadar kortisol dengan enzyme-linked immunosorbent assay (ELISA). Rata-rata kortisol sapi betina sebelum vs setelah restraint pada K1 dan K2 masing-masing adalah 55,64±52,13 vs 21,23±20,63 ng/ml dan 127,48±150,19 vs 106,28+75,39 ng/ml. Rata-rata kortisol sapi jantan sebelum vs setelah restraint pada K1 dan K2 masing-masing adalah 40,28±38,12 vs 24,21±17,21 ng/ml dan 49,51±38,67 vs 69,62±63,98 ng/ml. Rata-rata kadar kortisol tanpa memperhatikan jenis kelamin pada K1 dan K2 masing-masing adalah 79,50±88,50 vs 95,80±69,69 ng/ml (P>0,05). Disimpulkan bahwa tidak terdapat perbedaan kadar kortisol sebagai indikator stres pada sapi-sapi yang dipotong di RPH Yogyakarta yang mengalami restraint dengan restraining box tipe Mark 1 dan restraint metode konvensional. Penelitian ini menggunakanRancangan Acak Lengkap (RAL) pola faktorial dengan 2 faktor yaitu faktor Aadalah jenis susu yaitu a1 = Susu Sapi, a2 = Susu Kambing, a3 = Susu Bubuk. faktor Badalah lama Penyimpanan b1 = 0 hari, b2 = 7 hari, b3 = 14 hari. Parameter yangdiamati pada penelitian ini adalah kadar asam laktat, dan total mokroba. Hasilpenelitian menunjukkan bahwa kadar asam laktat yoghurt berkisar antara 0,74 - 1,68% dengan rataan sebesar 1,18 %. Selanjutnya terlihat bahwa jumlah bakteri asamlaktat tertinggi (1,68%) diperoleh pada susu kambing dengan lama penyimpananbeku (7 hari). Sedangkan jumlah bakteri asam laktat yang terendah (0,74%)diperoleh pada susu bubuk lama penyimpanan beku (0 hari). The research was carried on 9 district of Banda Aceh city which aimed to get the knowledge from acehness people about medicinal effect of euphorbiaceae plant such as: Euphorbia pulcherrima, Euphorbia hirta, Phyllanthus urinaria, Ricinus communis, Aleurites molucccana, Manihot esculenta, Jatrophe curcas, Codiaeum variegatum, dan Hevea brasilensis. The method used was PRA (Participatory Rural Appraisal) which purposive sampling for respondences: keuchik, tuha peut, dan tabib on every villages. The parameters of this research was: the kinds of disease, organ of plant used, mixing way of medicine, and how to use. The result showed that from 455 respondents, all of them, have the indigenous knowledge about Jatropha curcas as medicinal plant. Contrary of Euphorbia pulcherrima, Manihot esculenta, Codiaeum variegatum, and Hevea brasilensis are unknown as medicinal plant. Phyllanthus urinaria is the most species used, because can heal 10 disease. Metode penelitian ini menggunakan metode survei dengan pengambilan data secara sampel dengan cara Purposive Random Sampling. Penelitian ini menetapakan 80 responden dari peternak di empat kecamatan, dan 10 responden dari pemotong hewan atau pedagang daging di empat kecamatan untuk menjawab kuesioner yang telah disediakan. Dari hasil kajian diperoleh, populasi sapi potong; perbandingan persentase antara sapi jantan dan betina yaitu 35 persen berbanding 65 persen. Sistem pemeliharaan; 76 persen peternak menggunakan sistem semi intensif, selebihnya peternak menggunakan sistem ekstensif dan intensif. Sistem pengadaan bibit dan sistem perkawinan; 92 persen peternak memperoleh bibit dari induknya, sementara itu peternak yang menerapkan sistem perkawinan secara alami yaitu sebanyak 91 persen, hanya 1 persen peternak yang menerapkan kawin buatan (Inseminasi Buatan). Jumlah pemotongan; jumlah pemotongan sapi yaitu sebanyak 6.136 ekor sapi setiap tahunnya. Asal sapi yang diperoleh; sebanyak 100 persen sapi diperoleh dari dalam daerah ketika pemotongan pada hari biasa, 20 persen sapi didatangkan dari luar daerah ketika hari peukan serta 80 persen sapi didatangkan dari luar daerah ketika hari besar Islam (meugang). Konsumsi daging sapi; produksi daging sapi yaitu sebanyak 528 ton per tahunnya, sementara itu kebutuhan konsumsi daging yaitu 834 ton per tahun, artinya Kabupaten Pidie masih kekurangan sebanyak 306 ton daging sapi setiap tahunnya. Hasil penelitian data dengan menggunakan metode Net Present Value (NPV) pada tingkat bunga 13 persen per tahun, hasilnya positif Rp. 814.360,- kemudian dengan menggunakan metode Internal Rate of Return (IRR) hasilnya yaitu 35,92 persen, selanjutnya dengan menggunakan Benefit Cost Ratio (B/C Ratio) hasilnya 1,45 persen. Ini menunjukkan bahwa kegiatan daur ulang sampah sangat layak secara ekonomi. Berdasarkan hasil penelitian diharapkan agar pemerintah maupun masyarakat lebih mengembangkan kegiatan ini agar sampah di Kota Banda Aceh dapat diatasi secara efektif dan efisien.
