The objective is to know the ability of yeast to inhibit the growth of pathogenic bacteria in milk. They were Candida curiosa, Brettanomyces custersii, and Kluyveromices lactis. Hasil maserasi bunga kamboja putih dengan pelarut n-heksana menghasilkan ekstrak bewarna kuning dengan berat sebesar 21,06 g dengan rendemen ekstrak yaitu 2,78 %. Ekstrak n-heksana dipisahkan dengan Kromatografi Kolom Cair Vakum (KKCV) sehingga diperoleh 7 kelompok fraksi gabungan yaitu fraksi A (0,38 g), fraksi B (0,31 g), fraksi C (5,83 g), fraksi D (3,83 g), fraksi E (1,92 g), fraksi F (1,42 g), dan fraksi G (0,3 g). Ekstrak kasar dan fraksi-fraksi yang diperoleh diuji aktivitas antibakteri terhadap bakteri S. aureus dan E.coli dengan metode Kirby-Bauer menggunakan cakram dengan konsentrasi 1%, 10%, 20%, kontrol positif (kloramfenicol), dan kontrol negatif (n-heksana). Hasil uji aktivitas antimikrobial menunjukkan bahwa fraksi G memiliki potensi sebagai antimikrobial terhadap bakteri S.aureus dan E.coli dengan nilai zona hambat masing-masing pada kosentrasi 20% adalah 14,2 mm dan 9,1 mm. The study was conducted in two stages. The first stage included sampel preparation, phytochemical test, λ max determination, testing of antioxidant activity by reducing power method, and TLC analysis. In stage 2 purification using column chromatography, phytochemical test, and antioxidant activity test were performed. The sample was dissolved by using 70% ethanol and 1.5 N HCl with a ratio of 85:15 (v/v). Phytochemical test results showed that the sample consisted of not only flavonoids but also tannins. The Rf value for both sample and standard are respectively 0.55 and 0.61 with absorbance value of 0.826 and 0.845. The sample was then purified using column chromatography and 25 fractions were grouped based on the color intensity abbreviated as FA, FB and FC. Photochemical test was then conducted to examine the purity of obtained fraction and FB was found to be relatively purer than other fractions. Reducing power test of FB with variation of weight 10 mg, 20 mg, 30 mg, 40 mg, and 50 mg gave the percentage inhibition 64.1, 6.7, 67.9, 70.3 and 73.6, respectively, while the percentage value of α- tocopherol at simmilar weight variation are 32.1, 36.7, 43.5 and 50.2%. Klebsiella pneumoniae merupakan bakteri yang dapat menghasilkan Extended Spectrum Beta Lactamase (ESBL) yang menyebabkan bakteri tersebut resisten terhadap beberapa golongan antibiotik. Munculnya resistensi dan berbagai efek samping dari antibiotik menuntut penemuan dan pengembangan bahan alam sebagai antibakteri. Salah satu bahan alam yang dapat dijadikan sebagai antibakteri adalah tanaman daun kumis kucing (Orthosiphon stamineus Benth.). Tujuan penelitian adalah untuk mengetahui aktivitas ekstrak etanol daun kumis kucing (Orthosiphon stamineus Benth.) terhadap Klebsiella pneumonia dan mengetahui pengaruh peningkatan konsentrasi ekstrak etanol daun kumis kucing (Orthosiphon stamineus Benth.) terhadap luas zona hambat pertumbuhan K. pneumoniae. Penelitian ini menggunakan Rancangan Acak Lengkap yang terdiri atas 4 kelompok perlakuan dan 1 kelompok kontrol negatif dengan 5 kali pengulangan. Kelompok perlakuan terdiri atas konsentrasi ekstrak etanol daun kumis kucing 25%, 50%, 75%, dan 100%, sedangkan kelompok kontrol negatif adalah larutan carboxy methyl cellulose (CMC) 1%. Uji aktivitas anti bakteri dilakukan dengan metode difusi cakram Kirby-Bauer. Hasil pengamatan menunjukkan konsentrasi ekstrak etanol daun kumis kucing 25%, 50 %, 75%, dan 100% masing-masing memiliki zona hambat rata-rata 12,2 mm, 13 mm, 13.8 mm dan 15,8 mm. Berdasarkan analisis data dengan ANOVA menunjukan ekstrak etanol daun kumis kucing (Orthosiphon stamineus Benth.) memiliki pengaruh yang nyata dalam menghambat pertumbuhan Klebsiella pneumoniae.
