In this study, optimized the physical parameter of SSF in a shallow aluminium tray (30 cm x 20 cm x 6 cm) was carried out  in order to maximize  reducing sugar production by A. niger USM AI1 using oil palm frond as a substrate.  Production of reducing sugars before optimization in tray system was 185.00 ± 2.85 mg / g substrate with the fungal growth of 2.32 mg ± 0.06 glucosamine / g substrate. However, after optimization of physical parameter (amount of substrate, inoculum size, temperature and moisture content in substrate) the maximum reducing sugar yield was achieved 251.13 ± 1.95 mg / g substrate with the fungus growth of 3.21 ± 0.10 mg glucosamine / g on the 4 days of cultivation period.  The results showed that the production of reducing sugar was increased by 35.7% compared to before optimization.   Thus, optimized the physical parameter of solid state fermentation in a tray system has succeeded in increasing the production of reducing sugars. The clay is commercially available and named as “kieserite” produced by local companies. The sample was taken randomly from fertilizer shops and analysed with AAS and XRF methods. XRF data of local kieserite showed the dominated chemical content; MgO(33.19%); SiO2(33.11%); CaO(1.90%);Al2O3 (5.77%) and Fe2O3(6.32%). The MgO content is higher than that of magnesium sulphate monohydrate (CAS No.: 14168-73-1) which is only 28%. The concentration of soluble magnesium ions of local kieserite naturally soluble was 0.0186% which is very low compared with the soluble magnesium of magnesium sulphate monohydrate (CAS No.:14168-73-1) which is 25%. The Enhancing the magnesium solubility was done by soaking the local kieseriteinto 0.1-0.5M HNO3 before neutralized with NH4OH or mixing the local kieserite with in alluvial, entisol, red-yellow-podsolik (RYP) soils with and without addition of 0.1-0.5M of (NH4)2SO4. The amount of dissolved magnesium of kieserite was found morein entisol soil than the other type of soils. The concentration increased up between 0.59-1.00% along with the incrementof (NH4)2SO4 concentration (0.1-0.5 M). A study was carried out to partial characterize enzyme from exretory/secretory of A. galli L3 stage based on influence of inhibitor. A. galli L3 were recovered from intestines of 100 heads chickens 7 days after oesophagus inoculation with 6000 L2. L3 recovered in this manner were cultured (5 – 10 ml-1) in flasks containing rosswell park memorial institute (RPMI) 1640 media, pH 6.8, without phenol red. Cultures were incubated at 370C in 5% CO2 and culture fluid was collected after 3 days in culture. Excretory/secretory was prepared from metabolic product of L3 released in culture medium. The enzyme activity was assayed against casein 2%. The amount of concentration was determined from the absorbance at 595 nm. Inhibitor sensitivity on enzyme activity was studied in phenil methanyl methane sulfonyl fluoride (PMSF) 0,5 and 1 mM.    The result showed that enzyme activity of A. galli larvae was inhibited by PMSF 0.5 and 1 mM, which enzyme activity remainded 1,8 and 0%, respectively.
