Tujuan dari penelitian ini adalah untuk mengetahui pengaruh kepemimpinan dan budaya organisasi terhadap kepuasan kerja baik secara parsial maupun simultan. Selain itu juga untuk melihat pengaruh kepemimpinan dan budaya organisasi terhadap kinerja baik secara langsung maupun tidak langsung. Adapun yang menjadi objek penelitian ini adalah pengaruh kepemimpinan dan budaya organisasi terhadap kepuasan kerja dan dampaknya pada kinerja PNS setdakab Abdya, dengan jumlah responden sebanyak 146 orang. Sapi yang digunakan dalam penelitian ini adalah dua puluh ekor sapi perah peranakan Friesian Holstein, berumur 3-8 tahun sudah pernah beranak minimal satu kali, mempunyai siklus reproduksi normal, dan kondisi tubuh sehat. Seluruh sapi dibagi ke dalam dua kelompok yang masing-masing terdiri atas 10 ekor sapi. Kelompok I adalah sapi fertil sedangkan kelompok II adalah sapi yang telah diinseminasi lebih dari tiga kali tetapi belum atau tidak bunting. Profil biokimia darah yang diukur adalah kadar total protein, total kolesterol, glukosa, dan kalsium. Data dianalisis menggunakan t-test. Hasil penelitian menunjukkan bahwa total protein; total kolesterol; glukosa; dan kalsium pada kelompok I vs kelompok II masing-masing adalah 6,77±0,38 vs 6,82±0,821 g/dl (P>0,05); 166,08±37,06 vs 125,95±38,108 mg/dl (P<0,05); 68,40±9,60 vs 48,58±6,675 mg/dl (P<0,01); dan 9,90±1,43 vs 9,23±0,94 mg/dl (P>0,05). Disimpulkan bahwa sapi-sapi perah yang mengalami kawin berulang mempunyai total kolesterol dan kadar glukosa yang lebih rendah dibanding sapi-sapi perah yang fertil. Tiga sampel darah kuda Sandel Wood dari Kabupaten Sumba Timur digunakan untuk isolasi DNA. Produk isolasi DNA diamplifikasi pada daerah D-loop parsial mitokondria dengan metode PCR, primer ECLOOP forward 5’-CGCACATTACCCTGGTCTTG-3’ dan primer ECLOOP reverse 5’GAACCAGATGCCAGGTATG-3’, pada kondisi predenaturasi 94° C selama 5 menit, denaturasi 94° C selama 30 detik, annealing 56° C selama 30 detik, elongation 72° C selama 45 detik, dan post elongation 72° C selama 5 menit sebanyak 35 siklus. Reaksi polymerase chain reaction (PCR) menghasilkan produk sepanjang 492 bp kemudian dilakukan pengurutan. Produk pengurutan D-loop dibandingkan dengan spesies kuda lain dari Genbank dan dianalisis menggunakan MEGA versi 4.1. Hasil analisis diperoleh 66 situs nukleotida yang berbeda. Urutan nukleotida D-loop yang dianalisis menggunakan Kimura 2- parameter ditemukan nilai paling kecil 0% dan nilai paling besar 2,9%. Konstruksi pohon filogenetika menggunakan metode neighbour joining dengan nilai bootstrap 1000 kali menunjukkan hubungan kekerabatan yang sangat dekat di antara kuda Sandel Wood. Kultur T. hamatum WSY-03 menggunakan dioptimasi metode komposit terpusat (Central Composite Design/CCD) dan uji aktivitas enzim menggunakan metode Filter Paperase Unit (FPU). Data dianalisis dengan metode respon permukaan (Respon Surface Methodology/RSM) menggunakan Software Design Expert 8.0. Kondisi optimal aktivitas selulase T. hamatum WSY-03 adalah pada komposisi substrat dedak padi 100 g, kelapa (100 ml), urea (2 g) dan bibit Trichoderma (2 g) dengan nilai FPU 13 U/ml.Kata kunci: Selulase, Trichoderma hamatum WSY-03, metode komposit terpusat, metode Filter Paperase Unit (FPU), metode respon permukaan (RSM).ABSTRACTThis study aims to obtain the optimum condition of cellulase activity. This research was conducted in Biology Cell Molecular Laboratory and Microbiology Laboratory, Department of Biology, Mathematics and Natural Sciences Faculty, Syiah Kuala University from August to October 2013.The substrate used rice husk is added coconut water, urea and inoculum Trichoderma hamatum WSY-03. Culture T. hamatum optimaze using Central Composite Design (CCD) and enzyme activity assay using Filter Paperase Unit (FPU). Data was analyzed using Response Surface Methodology (RSM) with Design Expert 8.0 software. The optimum cellulase activity in the composition 100 g rice husk for 1 week within consentration coconut water (100 ml), urea (2 g) and T. hamatum WSY-03 (2 g) with level FPU 13 U/mL.
