Pengambilan sampel dilakukan pada Maret-April 2014 di empat Tempat Pendaratan Ikan (TPI), yaitu TPI padang Seurahet, TPI LabuhanHaji, TPI Idi Rayeuk dan PPS Lampulo. Sebanyak 1cm2jaringan sirip dikumpulkandari individu hiu kemudian diawetkan dengan 96% etanol, dilanjutkan proses ekstraksi kemudian diikuti proses amplifikasi dan sequensing. Hasil penelitian menunjukkan sebanyak 7 spesies hiu berhasil diamplifikasi dari 67 sampel yang dikoleksi dengan lokus Cytochrome Oxidase Subunit I (COI) dan panjang DNA 600-654 bp. 7 spesies hiu tersebut terdiri dari: Carcharinus sorrah, Carcharinus albimarginatus, Carcharinus amblyrinchos, Carcharinus falciformis, Triaenodon obesus, Sphyrna lewini dan Centrophorus granulosus. Analisis maximum likelihoodmenunjukkan 67 sampel tersebut membentuk pohon filogenetik yang membagi 8 kelompok percabangan termasuk outgroup. Taxonomi berdasarkan DNA barcodingselaras dengan taxonomi berdasarkan morfologi yang telah ada, oleh karena itu penelitian ini telah berhasil menvalidasi status taxonomi 7 spesies hiu di Perairan AcehKata kunci: Aceh, Ikan hiu, DNA Barcoding, Cytochrome Oxidase Subunit I (COI).
