Date A point or period of time associated with an event in the lifecycle of the resource 2012-11-30 Description An account of the resource DNA barcoding has been accepted nowadays as a globally accessible tool to delimitate and identify new species, as it can provide accurate and automated species identification through the use of standard gene, Cytochrome oxidase I (COI). As a result, DNA barcoding has become a perfect molecular tool to identify fish products and detect wrongly labelled fish in the market as it can identify fish species even without the presenceof complete morphological characteristics. Here we collected samples from sushi restaurants to investigate and identify the precise species identity of raw fish sushi products because most of them were labelled with only common names such as Tuna, Salmon and Butterfish without providing the scientific name of the fishes. Species identities of seven specimens from two sushi restaurants were DNA barcoded utilising the cytochrome oxidase I (COI) gene. The data were BLAST with GenBank and BOLD to confirm species identification. The results showed 100% matches that there was nosubstitution and wrongly labelling of species as the Salmon sushi matched the database sequences of Salmo salar, Tuna to Thunnus albacares and Butterfish to Lepidocybium flavobrunneum. The bioetanol development from biomass bases on lignocellulose like pulp of coffee is one of alternative energy which has potential to be applied in Indonesia. Beside of raw material sources that are so many in our country, the process is also environmentally friendly. Conversion of coffee pulp becomes ethanol using Simultaneous Sacharification and Fermentation (SSF) technology by cellulase had been done on this research. Sacharification process or hydrolysis process, cellulase enzyme will break cellulose polymer becomes glucose. Then, glucose through fermentation is changed to ethanol by using yeast Saccharomyces cerevisiae. This study using Completely Randomized Design (CRD) with factorial pattern consisting of two factors. Factor I is the optimum pH (P) consists of three standards are: P1 = 4, P2 = 4.5, P3 = 5. Factor II is the incubation time (fermentation) (T) consisting of four standard that is T1 = 24 hours, T2 = 48 hours, T3 = 72 hours, and T4 = 96 hours. Pretreatment to break the lignin is done by soaking in 2% H2SO4 for 20 hours in the erlenmeyer. The analysis performed consists of the initial analysis and final analysis. Initial analysis consisted of analysis of water content, pH analysis, levels of cellulose and glucose levels, against the skin of the coffee pulp before the simultaneous saccharification and fermentation (SSF), whereas the final analysis included analysis of ethanol content, the analysis of cellulose and glucose from a solution of ethanol produced. The optimum pH treatment (P), incubation time (fermentation) (T) and interaction optimum pH and incubation time (fermentation) (PT) has very significant (P ≤ 0.01), on levels of ethanol produced. The highest levels of ethanol obtained by fermentation of 96 hours and the optimum pH 4.5 and 5. Hemp offers characteristics that are similar to wood properties. In line with this research, three-layered particleboards under utilization of hemp material were developed. The hemp was used in combination with industrial produced particles to find out which amount of wood can be substituted by hemp and further to investigate which decreased densities can be realized by the use of hemp. Hemp can be used for the production of lightweight particleboards due to its lower bulk density compared to wood. All developed hemp/wood based particleboards were produced under industrial parameters in laboratory and pilot scale using conventional urea formaldehyde-resin, hardeners, and hydrophobic substances. The mechanical technological properties of all produced innovative particleboards and reference particleboards were determined according to current DIN/EN standards. In detail density profile, bending strength, internal bond strength, surface strength, and thickness swelling were determined. Compared to the produced reference particleboards, consisting only of wood particles, the developed hemp/wood particleboards possessed better mechanical-technological properties. Furthermore it was realized to produce particleboards with densities of 550 kg/m³ by using hemp due to its low bulk density which fulfills the given DIN/EN requirements.
