Empat strain telah diuji potensinya untuk menghasilkan lipase yaitu PLS A, PLS 75, PLS 76 dan PLS 80. PLS 80 mampu menghasilkan lipase setelah diinkubasi pada suhu 70oC selama 24 jam, pH 9 dalam media produksi agar yang mengandung minyak zaitun, tween 80 dan rhodamin B. Aktivitas maksimum lipase tercapai pada jam ke-16 inkubasi, pH 9, suhu 70oC dengan kecepatan pengadukan 150 rpm. Aktivitas yang diperoleh sebesar 18,05 U/mL. PLS 80 merupakan mikroorganisme Gram negatif. Analisis morfologi sel menggunakan SEM menunjukkan PLS 80 berbentuk batang yang panjang yang merupakan ciri Geobacillus sp. Pohon filogenetik dibentuk untuk mengetahui kekerabatannya dengan mikroorganisme lain berdasarkan hasil sekuensing gen 16S rRNA. PLS 80 memiliki kemiripan 99% dan terletak dalam cabang yang sama dengan G. kaustophillus strain Da2. Peubah yangdiamati adalah indek vigor, panjang plumula, panjang radikula, rasio panjangradikula per panjang plumula dan beratkering. Sebanyak 60 ekor ayam petelur berumur 22-44 bulan yang telah mengalami penurunan produksi sekitar 50-60% asal peternakan ayan petelur Jantho Farm, Aceh Besar digunakan dalam penelitian ini. Seluruh ayam petelur ditempatkan dalam kandang batere yang dibagi ke dalam 6 kelompok perlakuan, masing-masing perlakuan terdiri dari 10 ekor ayam petelur. Perlakuan I diinjeksi dengan 0,1 ml NaCl fisiologis, perlakuan II diinjeksi dengan 15 IU PMSG, perlakuan III diinjeksi dengan 0,1 ml ekstrak hipofisa sapi, perlakuan IV diinjeksi dengan 0,2 ml ekstrak hipofisa sapi, perlakuan V diinjeksi dengan 0,3 ml ekstrak hipofisa sapi, dan perlakuan VI diinjeksi dengan 0,4 ml ekstrak hipofisa sapi. Injeksi dilakukan secara intramuskular pada otot dada setiap dua minggu sekali dan diamati selama 42 hari. Data hasil penelitian dianalisis secara deskriptif. Hasil penelitian menunjukkan total rata-rata berat dan ketebalan cangkang telur tertinggi terdapat pada kelompok perlakuan V yakni masing-masing 66,20+1,67 g dan 0,43+0,02 mm, sedangkan total rata-rata berat dan ketebalan cangkang telur terendah terdapat pada kelompok perlakuan I yakni masing-masing 63,66+1,86 g dan 0,416+0,03 mm. Dosis optimal ekstrak hipofisa sapi dalam mempertahankan berat dan ketebalan cangkang telur ayam petelur pada fase akhir produksi adalah 0,3 ml.
