<text>Microsatellite analysis in biotechnology of animal reproduction (A Review)ABSTRACT. In year 1970 was found nucleotide sequence which have repeated sequence of nucleotide. with high polymorh and using PCR could be amplified. That sequenz of nucleotide called Microsatelite. Microsatelite consist of 1 – 6 repeated nucleotide, which is CA repeated as mostly a repeated DNA in the animal (Tautz and Renz, 1984). Based on difference of long and amount of repeated nucleotide, there are three kind of DNA satelite, midi-, mini- and microsatelite (Matiat and Vergmaud, 1982). Microsatelite analysis was used to analyze of paternity and identity of animal, which was done as a conventional analysis with blood group analysis. The advantage of microsatelite analysis compare to blood group system are the exclution probability was high (EXP 99.9%), needs small sampel (tissues, sperm or follicel of hair), could be use for all animal without special age and possible for died animal.</text>

The objective of this research was to know the effect of enzim phytase supplementation on relative organ digestion broiler. The research was done for weeks at Animal Fees and Nutrition, Faculty of Agriculture, Sriwijaya University in Mei to Juni 2008 utilized. A Completely Randomized Design with 6 treatments {P0 (100% basal diet), P1 (100% basal diet + 500 FTU/kg), P2 (100% basal diet + 600 FTU/kg), P3 (100% basal diet + 700 FTU/kg), P4 (100% basal diet + 800 FTU/kg),P5 (100% basal diet + 900 FTU/kg)}. Each treatments was replicated for 3 times. The parameters are final body weight, (%) proventrikulus, (%) relatif intestine, (%) pancreas and (%) liver weigh broiler chickens . The research showed that adding enzim phytase has join no significant effect (P>0.05) on the body, liver, proventrikulus, ventrikulus weight, bur it showed significant effect on relative intestines and pancreas weight (P>0.05) of broiler chickens . the average weight of body weight was 1117.50-1873.00 gram, proventrikulus was 0,44-0,64% ventrikulus was 1.30-2.32%, intestine was 2.74-4.39%, pancreas was 0,21-0,32% and liver was 2,35-3.01%.</text>

<text>The quality and palatibility of silage and hay complete ration based on organic primer garbage in peranakan etawah (PE) poadABSTRACT. This research was conducted to study the quality and palatability of silage and hay complete ration based on organic primer garbage in nine heads of Peranakan Etawah (PE) Goat. The quality of silage and hay was evaluated By measuring pH, total number of lactic acid Bacteria (LAB), palatability, dry matter (DM) and organic matter (OM) aroma and color of silage. Nine heads of peranakan etawah were grouped based on their lactation periode and randomly assigned to one of tree dietary treatments. The treatments were (1) Control rations; (2) silage complete ration (SRK); and (3) Hay complete ration (HRK). The results indicated that the silage had PH 4.15 and total number of LAB was 2.4 x 10 cfu/g of silage. Dry matter and organic matter digestibility of SRK were better than those of HRK. The palatability of SRK was higher than HRK but less than control. Storage of SRK for did not change dry matter and organic matter content. Organoleptic analysis indicated that texture and color of HRK were changed at after storage, while SRK was still constant. It is concluded that silage composed of organic primer garbage has high quality in terms of physical, chemical and microbial characteristics, prolong storage.</text>

<text>Purification yolk immunoglobulin of hens vaccinated against excretory/secretory Ascaridia galli L3 larvae stageABSTRACT. The main immunoglobulin fraction of poultry is called IgY, in order to distinguish it from the mammalian IgG. This article focus on purification yolk immunoglobulin of hens vaccinated against excretory/secretory Ascaridia galli larvae to obtained purity IgY. Active vaccinations with excretory/secretory antigen were applied intra muscularly of chickens with an initial dose of 80 μg. The vaccinations were repeated three times with dose of each 60 μg with an interval of one week. The first vaccinations were excretory/secretory antigen mixed with Fruend Adjuvant Complete and subsequently mixed with Freund Adjuvant Incomplete. Antibody response in yolk was detected at weekly intervals by agar gel precipitation test (AGPT). The chicken’s eggs were collected from 49 day after vaccinations. IgY was extracted from egg yolks by means of ammonium sulphate and purified using fast performance liquid chromatography (FPLC). The purity of anti-ekscretory/secretory IgY protein was determined by Bradford method (λ = 280 nm). The result showed that antibody in yolk was begun detect with AGPT at four weeks after vaccination. IgY concentration after purification was 0,875 ± 0.251 mg/ml. This study has shown that the product released in vitro by L3 stage A. galli is capable of stimulating humoral immunity by mean of producing antibody through yolk as biologic manufacturer could be a good choice.</text>

<text>Rumen metabolism product on growing cattle ongole gradeABSTRACT. The utilization of fermented fibrous waste material had improved the product of rumen metabolism in vitro, nevertheless, it was not always the same result if treated on animal, in vivo. An experiment had been conducted to investigate the result of the in vitro trial to the growing male cattle of Ongole Grade. The experiment had been conducted by experimental method using twelve (12) growing male cattle of Ongole Grade. The trial was conducted in Completely Randomized Block Design, during seven months period, three months adaption, two weeks preliminary, three months feeding trial and two weeks digestion trial. The treatment tested were: R0 = control ration, non fermented; R1 = mesh type fermented and R2 = pellet type fermented. The variables measured were: the production of Total Volatile Fatty Acid (T-VFA); asetate (C2), propionate (C3), butyrate (C4). The result shown that the treatment, biological and physical, of complete feed, does not influence on the production of T-VFA as well as on the composition (C2, C3 and C4). Nevertheless based on all the variables measured, the biological treatment with fermentation tends to increase the production of rumen metabolism, and the highest result of the products of T-VFA = 112,43 + 17,2 mM/L, C2 = 40,46 + 8,98 mM/L, C3 = 34,57 + 7,35 mM/L, C4 = 31,17 + 7,35 mM/L, were resulted from pellet type of complete feed. The result of experiment concludes : (1) based on the production of VFA as well as its composition, and the balance of C2 / C3, the utilization of complete feed both mesh and pellet type increases the efficiency of energy utilization on the growing male of Ongole Grade Cattle; (2) based on rumen metabolism product, the recommended complete feed for growing male cattle of Ongole grade is in the form of fermented pellet.</text>

This study aims to examine the energy sources of feed ingredients that can increase the production of Volatile Fatty Acids (VFA), N-NH3, microbial protein synthesis, total gas production and metabolic energy. The material used is as a source of rumen fluid inoculum from Frisian Holstein cows (FH) females, amoniasi rice straw, salt, mineral mix brand "Ultra Minerals' production Eka Farma Semarang, onggok wet and dry, corn, and rice bran. Observed variable is the concentration of (VFA), N-NH3, rumen microbial protein synthesis, and total gas production. Based on the analysis of diversity seen any significant effect (P<0.05) on total VFA concentration, N-NH3 and total gas but had no effect (P>0.05) on microbial protein synthesis. Conclusion of research is the provision of energy sources with rice bran treatment, onggok wet and dry corn flour can be used as fermentable carbohydrates on feed hay amoniasi in vitro.</text>

The purpose of this study was learn about the influence added lignosulfonate and bentonite by some processes production on physical characteristics of pellet form ration. The experimental design had used a Randomized Completely Design with 4 x 3 factorial and 3 replocats. Factor A were processes production, i.e : A0 = without processing, A1 = added 5% water spraying, A2 = added 5% hot water spraying and A3 = steam 45 minute on 1000C and pressure 1.7-1.8 kg/cm2. Factor R were added binder, i.e: R0 = control ration, R1 = control ration + 1.25% lignosulfonate and R2 = control ration + 2.5% bentonite. The parameters observed were water content, specific weght, specific density, packed specific density, angle of repose, particle size, water activity and durability of pellet. Result from this study showed some steam process by autoclave 45 minute and added binder lignosulfonate and bentonite was obviously influence specific weight, specific density, packed specific density, angel of repose, particle size, water activity and durability of pellet by percentage was 0.67 cm, 0.68 g/cm3, 0.75 g/cm3, 1.57 g/cm3, 26.990, even the water activity 0.81 wasn’t good. Added binder lignosulfonate and bentonite was obviously particle size was 0.67 cm and durability of pellet was 99.99%. Added lignosulfonate and bentonite by some process given the best on durability if it compared with commercials ration. The purpose of this study was to learn about the effect of using binder lignosulfonate and bentonite during six weeks period. The experimental design used a Randomized Completely Design with 3 x 4 factorial and 3 replications. Factor B was storage term (0. 2. 4 and 6 weeks). The parameters observed were water content, water activity factor higroskopic, particle size, specific density, bulk density, packed bulk density and pellet durability. The storage term very significant (P<0.01) influenced the water content and increased the water activity, decreased the factor higroskopic, particle size, specific density, bulk density, packed bulk density and durability of pellet. The lowest water content was 10.82% in 6 weeks. The lowest water activity was 0.74 in 0 week. The highest particle size was 0.669 cm in 0 week. The highest durability of pellet was 99.95% in 0 week. The highest specific weight was 1.30 g/cm3 in 2 weeks. The highest specific density was 0.738 g/cm3 in 0 week. The highest packed density was 0.738 g/cm3 in 0 week and the lowest factor higroskopic was 0.79% in 2 weeks.</text>

<text>(The evaluation of nutritive value and In Vitro digestibility of oil palm fronds through fermentation by using Aspergillus niger with different soluble carbohydrate sources) ABSTRACT. Oil palm frond (OPF) is one of potential sources of alternative feed, but has limited use due to high crude fiber and low crude protein contents. Fermentation is one of the methods widely applied to improve nutritive value of animal feed. The purpose of this research is to increase nutritive value of fermented oil palm fronds by adding different soluble carbohydrate source into fermentation media. The results of the experiments indicated that fermented oil palm fronds by Aspergillus niger had a significant effect (P<0,05) on the content of crude protein, crude fiber, and ash. Generally, fermented oil palm fronds with different soluble carbohydrate was able to increase the content of crude protein of oil palm fronds, but not optimal yet in reducing the crude fiber content of fermented substrate. However, the addition of rice bran on the fermentation medium showed the best results, characterized by increasing crude protein and decreasing crude fiber contents as well as improved dry matter and organic matter digestibility, reflected in high concentration of VFA. </text>

The aim of this research was to determine the ratio of birth sex of ettawa crossbreed goat by artificial insemination using spermatozoa swim up. This research was at Reproduction Laboratory Faculty of Veterinary medicine Syiah Kuala university and Livestock Group SMD, Bener Meriah Regency. Fifteen female goats with age 8-24 months where used in this study and divided into 3 treatment groups (P0, P1, P2) and 5 replications of each. The first group (P0) inseminated with sperm without swim up; P1 and P2 inseminated with sperm which swim up for 5 minutes (P1) and 10 minutes (P2). The result of conception rates observation and sex ratio of children born tabulated into tables. The data was analyzed by using analysis of variance (ANOVA) one way pattern followed by the multiple test Duncan. The results showed that the conception rate and birth sex ratio of male born had significantly different (P<0,05). The segregation of sperm which swim up increased the conception rate and birth sex ratio of male born.</text>
