Metode yang digunakan adalah metode pitfall trap, yang diletakkan pada habitat hutan yang memiliki ketinggian yang berbeda. Arthropoda yang diperoleh diindentifikasi sampai tingkat ordo, dan dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa pada kawasan hutan dengan ketinggian ± 1.500 dpl diperoleh 35 individu dari ordo Amphipoda, Araneae, Blattodea, Coleoptera, Collembola, Diptera, Homoptera, Hymenoptera, Lepidoptera, Orthoptera, pada ketinggian ± 1.600 dpl diperoleh 89 individu dari Ordo Amphipoda, Araneae, Coleoptera, Collembola, Diptera, Hymenoptera, Orthoptera, pinggiran telaga pada ketinggian ± 1.400 dpl diperoleh 29 individu dari Ordo Araneae, Coleoptera, Collembola, Diptera, Hymenoptera, Orthoptera, dan hutan dekat pondok dengan ketinggian ± 1.400 dpl diperoleh 26 individu dari Ordo Amphipoda, Collembola, Diptera, Hymenoptera, Orthoptera. The purpose of the research is to give evidence about how is the people’s behavior towards the wild life conservation area of Sibelia Mountain, the. The population of the research was all of patriarchs in three villages: Sawadai village, Tuokona and Gandasuli village. Furthermore, 46-47 people are chosen randomly from each village to get 140 patriarchs of total sample. Questionnaire instrument was used to collect the data. Questionnaire instrument and closed response interview guideline were used to collect the data of people’s behavior toward the wild life area conservation. Prior to distribute the instrument to the respondents, a test is carried out to the people who live in east Halmahera wild life area excluding from the research sample to get its validity and reliability. The result of analysis on people’s behavior in three different villages living in wild life area shows that they still have a low level of land utilization (about 20% - 25%). This result indicates that respondents who answered the questions of utilize of the conservation area tend to be in small number. This result also shows that the people of the three villages are having a tendency to do activities that caused environmental damage (contra conservation). The analysis result on people’s behavior in maintaining wild life conservation area of Sibela Mountain is also at a low level. From the three villages, the percentages are 8% - 13%. It indicates that people’s behavior tendency of maintaining or preserve the conserved area is at a very low level. Generally, the growth of G. erosa has negative allometric, isometric and  positive allometric fashion. This encourages the study of G. erosa living in the region of mangrove ecosystem of west-coast of Aceh Besar. The objectives of the research are to determine the growth pattern of shell length against body weight as well as the growth pattern of shell length against its total body weight. The research was conducted from February 2006 to January 2007 in the region of mangrove ecosystem of west-coast of Aceh Besar. The region was divided into three different locations. Each location was set a sample square with 10m x 10m in size which was perpendicular to coastal lines. There were 10 sub-squares also set in every sample square with 50cm x 50cm in size. Sample collection in the sub-square was carried out by scratching and disturbing the bottom of the waters in order to sieve them. G. erosa was then measured for its shell length, body weight, and total body weight. The growth pattern was analyzed by using Excel program and t-test. The result showed that a negative allometric growth pattern was found in the measurement of shell length against total body weight of G. erosa, whereas an isometric pattern was found of the measurement of shell length against its body weight. Moreover, a positive allometric pattern was figured out in the measurement of shell length against total body weight. The proliferation of shell length, body weight and total body weight has contributed approximately 58% to 59%. This indicates that the growth pattern of G. erosa in the region of mangrove ecosystem of west-coast of Aceh Besar has negative allometric, isometric and positive allometric fashion. Therefore, the study of the zones of both activities, hatching and nursery, must be done.  The study was aimed to determine the zonation of hatching of Gelonia erosa, indicators for hatching and nursery zones, and strategies of management of both zones in the brakhish area Leupung. Data was collected in three locations; Location I (River Leupung), Location II ( River Reuleng), and Location III (River Lhok Kulam). The methods of collecting data were Jarnis Larva Collector used for larva collection, in-situ used to collect biophysic data, and sweep area and destructive methods used to collect the young and mother of the species. It was found that in downstream of Location I, dominated by Nypa fructicans, there were 24 larvas, 10 and 61 individuals of young and mother of Geloina erosa respectivelly. Location II dominated by Nypa fructicans  and Fimbristilys sp had 10, 14, and 49 individuals of larva, young, and adult of Geloina erosa whereas Location III dominated by fructicans  and Fimbristilys sp as well had 12 and 65 individuals of young and adult of Geloina erosa. It can be concluded that the zone of hatching can be determined by the presence of larva, young and adult of Geloina erosa with fully-grown of gonad while the determination of nursery zone is relied on the presence of young and male and female adults of Geloina erosa with well-matured gonads. Whole blood DNA was extracted according to the protocol of Qiamp DNA Blood Mini Kit (Qiagen), and used as template for PCR (Polymerase Chain Reaction) which was conducted according the protocol of Takara Ex TaqTM PCR Kit in a Perkin-Elmer Thermal Cycler 9600. PCR products were purified using a Qiaquick PCR Purification Kit (Qiagen, USA). The sequencing reaction for RDP1 was performed using BigDye v.3.1. Cycle Sequencing Kit (Applied Biosystems,USA) followed the protocol of ABI Prism 3100-Avant Genetic Analyzer (Applied Biosystems, USA). Alignment of RDP1 nucleotide sequences of megapodes and that are obtained from international DNA data base (DDBJ-DNA Data Bank of Japan) with accession number AF 394649 (RDP1) were conducted by using Clustal-X. Characteristics of RDP1 were analyzed by employing the DNA Sequence Polymorphism (DnaSP) version 3.51 and Program MEGA version 4. The analysis result demonstrated there were insertion and deletion on RDP1 of species of Megapodes in Indonesia, which resulting in a gap of 1-10 bp and was concentrated in nucleotide 407-568. Alignments between species of Megapodes  and two species of Galliformes showed more gaps, which were concentrated on nucleotides 480-708. Its function is still unclear. This is an encouragement to study the gene by disrupting the expression of the gene. This study was aimed to generate Arabidopsis thaliana possessing suppressed Atnda1 utilizing RNA interference.  Double stranded nda1-RNAi silencing was chosen for this project. The generation of the lines was started by creating specific primer for Atnda1. The creation of nda1-RNAi cassette used the Gateway cloning. We utilized Agrobacterium strain GV3101 as a mediator to transfer the cassette into the plants. Dipping the flower into Agrobactrium solution was done to transfer the nda1-RNAi cassette into wild type Arabidopsis. The seeds collected from the mutant plants were checked in selective media. Contamination was the daunting problem in the process of checking the plants. This was crucial to ensure that the insertion has been done successfully. However, selection of the mutants must be done before analysis of the function is carried on the mutants.
